Un nuevo metodo para el mantenimiento de bacterias fitopatogenas que atacan el cultivo de la caraota (Phaseolus vulgaris L.)

Se incubaron los cultivos en el lab. a 24 + 1 grados centigrados hasta que el medio se seco completamente (de 5-6 semanas). El cultivo seco fue removido con pinzas esteriles y cortado en pequenos pedazos con tijeras esteriles, luego molido asepticamente en un mortero hasta conseguir un polvo muy fino. Los cultivos en forma de polvo se incubaron en tubos de vidrio esteriles y protegidos de la luz a 24 + 1 grados centigrados. A varios intervalos muestras del polvo fueron suspendidas en bufer salino esteril y se cultivaron muestras de la suspension en los medios apropiados para comprobar su viabilidad. X. phaseoli, X. phaseoli var. fuscans y C. flaccumfaciens se recuperaron facilmente de polvo guardado 14 meses, a concn. superiores a 103 celulas/mg de polvo seco. Todos los aislamientos bacterianos conservados en polvo seco fueron patogenicos cuando se inocularon en plantas de frijol var. Red Kidney. Con otros aislamientos de bacterias fitopatogenas que se probaron, todos mantuvieron su viabilidad por lo menos 4 meses en la forma anteriormente descrita. (RA)Summary: Isolates of Corynebacterium flaccumfaciens, Xanthomonas phaseoli, and X. phaseoli var. fuscans were grown in plates of YCA (yeast extract, calcium carbonate, agar) and isolates of Pseudomonas phaseolicola and P. syringae in plates of King's Medium B. Cultures were incubated in the lab. at 24 + 1 degrees Celsius until the agar was completely dry (between 5-6 wk.). The dry agar was removed with sterile forceps, cut into small pieces with sterile scissors, and aseptically ground to a fine powder with mortar and pestle. The fine powder cultures were stored in sterile glass vials at 24 + 1 degree Celsius. At various intervals, samples of the powder were suspended in sterile buffered saline, and aliquots of the suspensions were transferred to the appropriate medium to test their viability. Isolates of X. phaseoli, X. phaseoli var. fuscans, and C. flaccumfaciens bacteria were easily recovered from powder stored 14 mo., at concn. greater than 103 cells/mg of powder. All bacterial isolates were pathogenic when inoculated to bean var. Red Kidney plants. Several other isolates of phytopathogenic bacteria maintained viability for at least 4 mo. when stored in a similar manner. (AS)
