Se transmitio virus del mosaico del enanismo del frijol (BDMV) mecanicamente y mediante mosca blanca (Bemisia tabaci) de un cv. de frijol susceptible a otro tambien susceptible. Dos especies de Sida, que se sospechaba constituian malezas deposito de BDMV, tambien mostraron hospedantes susceptibles en pruebas de transmision por mosca blanca. La microscopia electronica de explantos ultradelgados de tejido foliar de frijol y Sida sp. infestado de BDMV mostro la presencia de agregados de particulas similares al virus en las celulas del floema. Se aislaron particulas nucleoproteinicas cuasiisometricas (A260 280 nm = 1.4), principalmente en pares que median 20 x 33 nm, de plantas de frijol infestadas de BDMV. La nucleoproteina purificada infecto las plantas de frijol cv. Topcrop, induciendo sintomas caracteristicos de BDMV. La nucleoproteina infecciosa disociada genero una especie de proteina simple con una masa molecular de 27,500 daltones y una banda de acido nucleico simple de aprox. 2.6 kb. El acido nucleico purificado fue resistente a ribonucleasa pero sensible a desoxirribonucleasa I y a nucleasa S1, indicando la presencia de ADN monocatenario. El virus aislado se relaciono serologicamente con CAMV, BGMV, mosaico amarillo del frijol mungo y virus del mosaico dorado del tomate. Se propone la asignacion del BDMV al grupo de geminivirus. Bean dwarf mosaic virus (BDMV) was transmitted mechanically and by the whitefly Bemisia tabaci from and to susceptible bean (Phaseolus vulgaris) cultivars. Two species of Sida, suspected weed reservoirs of BDMV, were also shown to be susceptible hosts by whitefly-transmission tests. Electron microscopy of ultrathin sections of BDMV-infected bean and Sida sp. leaf tissue revealed the presence of viruslike particle aggregates in phloem cells. Quasi-isometric nucleoprotein particles (A260 280nm = 1.4), found predominantly in pairs measuring about 20 x 33 nm, were isolated from BDMV-infected bean plants. Purified nucleoprotein was infective to Topcrop bean plants, inducing characteristic bean dwarf mosaic symptoms. Dissociated infectious nucleoprotein yielded a single protein species of molecular mass 27,500 daltons and a single nucleic acid band of approximately 2,6 kb. Purified nucleic acid was resistant to RNase but sensitive to DNase I and nuclease S1, indicating the presence of single-stranded DNA. The isolated virus was serologically related to African cassava mosaic, bean golden mosaic, mung bean yellow mosaic, and tomato golden mosaic viruses. Bean dwarf mosaic virus is proposed to be a new member of the geminivirus group. (AS) |
