Comparison between Xanthomonas campestris pv. manihotis (ISPP List 1980) and X. campestris pv. cassavae (ISPP List 1980) by means of phenotypic, protein electrophoretic, DNA hybridization and phytopathological techniques

Se estudiaron cepas de Xanthomonas campestris pv. manihotis y X. campestris pv. cassavae, aisladas de plantas de yuca de diferente origen geografico, mediante el analisis numerico de 267 caracteristicas fenotipicas, la comparacion computarizada de electroforegramas en gel de proteinas solubles, las determinaciones G+C de porcentajes de mol, hibridizaciones ADN:ADN y pruebas de virulencia. X. campestris pv. manihotis y pv. cassavae constituyeron entidades biologicas separadas que podian diferenciarse entre si mediante 4 caracteristicas bioquimicas, sus sintomas en yuca, sus electroforegramas de proteina soluble y sus caracteristicas del ADN. Dentro de cada patovar no se hallo correlacion entre fitopatogenicidad, origen geografico y ano del aislamiento de las cepas, por una parte, y las propiedades bioquimicas, fisiologicas y electroforeticas proteinicas por la otra. Dos cepas amarillentas de Xanthomonas, CIAT 1164 y CIAT 1165, aisladas de la yuca en Colombia, fueron genetica y electroforeticamente similares a X. campestris pv. poinsettiicola, pero no pudieron infectar a Euphorbia pulcherrima. X. campestris pv. poinsettiicola estuvo geneticamente mas relacionado con el grupo de X. campestris pv. manihotis que con el grupo de X. campestris pv. cassavae. (RA-CIAT) spa
Xanthomonas campestris pv. manihotis and X. campestris pv. cassavae strains, isolated from cassava plants of different geographical origin, were studied by numerical analysis of 267 phenotypic features, computer-assisted comparison of gel electrophoregrams of soluble proteins, mol percentage G+C determinations, DNA:DNA hybridizations, and virulence tests. X. campestris pv. manihotis and pv. cassavae constituted separate biological entities which could be differentiated from each other by 4 biochemical features, their symptoms on cassava, their soluble protein electrophoregrams, and their DNA characteristics. Within each pathovar no correlation was found between phytopathogenicity, geographic origin, and year of isolation of the strains, on one hand, and the biochemical, physiological, and protein electrophoretic properties on the other. Two yellowish Xanthomonas strains, CIAT 1164 and CIAT 1165, isolated from cassava in Colombia were genetically and electrophoretically similar to X. campestris pv. poinsettiicola, but were unable to infect Euphorbia pulcherrima. X. campestris pv. poinsettiicola was genetically more related to the X. campestris pv. manihotis cluster than to the X. campestris pv. cassavae cluster. (AS) eng
