Screening and quarantine of cassava from East Africa

Se seleccionaron 248 estacas provenientes de 10 variedades de Kenya, 4 de Tanzania y varias de Guinea. Las plantas se mantuvieron en invernaderos y casas de malla a prueba de insectos y de temperatura controlada. Se seleccionaron en 3 sitios plantas sanas de yuca segun su reaccion a virus locales de ocurrencia natural. Se analizo la presencia de virus transmitidos por la savia, en hojas molidas, mediante uno de 2 metodos, una suspension de alumina y celite, 0,02M tampon fosfatico a pH 7, que contenia 0,005 M DIECA y Natioglicocolate y 0,001M EDTA. Se inocularon varias plantas con los extractos, se cortaron en tajadas delgadas y se examinaron por medio del microscopio electronico, siguiendo tecnicas corrientes, y por medio de procedimientos de tincion negativa. Las plantas cultivadas en casa de malla se desarrollaron mejor que las cultivadas en invernadero y en el campo. Se encontro el virus mosaico del tabaco (TMV) en la yuca pero no indujo sintomas. No se obtuvieron resultados definitivos respecto a la transmision de virus o a las causas del mosaico africano. Como resultado de este trabajo, numerosas variedades remitidas desde el este de Africa fueron declaradas no portadoras del virus, multiplicadas y enviadas a Guinea. Plants were kept in insect-proof, temperature-controlled glass and screenhouses. Healthy cassava plants were screened against naturally ocurring local viruses in 3 locations. Ground leaves were checked for the presence of sap- transmissible viruses in either a suspension of alumina and Celite or 0.02 M phosphate buffer at pH 7 containing 0.005 M DIECA and Nathioglycollate and 0,001 MEDTA. The extracts were inoculated in test plants and tested by electron microscopy of thin sections using standard techniques and by negative staining. Plants grown in screenhouses grew better than those in glasshouses and in the field. Tobacco mosaic virus was present in cassava but did not induce symptoms. No definite results were obtained on natural transmission of viruses or causes of cassava mosaic. As a result of this work, a no. of cassava varieties introduced from East Africa by the Institute Nazionale Piante da Legno were found virusfree, multiplied and sent to Guinea. (CIAT)
